ABSTRACT

In this study, black phosphorus nanosheets (BP) were prepared by the ordinary liquid phase method, and resveratrol was loaded on the BP after being modified by polyethylene glycol. The brain targeting of BP was investigated by fluorescent protein labeling, and the effects of black phosphorus on cerebral ischemia/reperfusion injury were studied by 2,3,5-triphenyltetrazolium chloride (TTC) staining, neurobehavioral evaluation, and brain edema. Protein immunoblotting analysis was used to explore the molecular mechanism of the BP drug delivery system on ischemic brain injury. Hemolysis test and hematoxylin-eosin (H&E) staining were used to evaluate its biocompatibility. The results showed that BP had excellent drug loading capacity, uniform drug loading system structure and particle size, stable drug release curve, and excellent photothermal effect. Through the analysis and comparison of fluorescence intensity, it was found that BP can increase the permeability of blood-brain barrier (BBB) under the condition of near-infrared light assisted irradiation, and make drugs more pass through the BBB. In addition, the black phosphorus nano tablet drug delivery system can significantly improve the neurobehavioral disorder of mice after modeling, and the cerebral infarction area and brain edema degree are significantly decreased. Western blot experiments showed that the drug delivery system could play an anti-ischemic brain injury role by activating the expression of antioxidant signaling pathway proteins nuclear factor E2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1). The hemolysis test and H&E test results of the BP drug carrier system showed that it had no obvious toxicity and high safety. In conclusion, the BP prepared in this study had high drug loading, good photothermal performance, and high safety. Under the near-infrared condition, they also have certain brain targeting ability, which can improve the therapeutic effect of drugs in the brain. Animal welfare and experimental procedures were following the regulations of the Animal Ethics Committee of the First Affiliated Hospital of Shihezi University.

ABSTRACT

The intestinal absorption properties of the main effective components (glycyrrhizic acid, isoliquiritigenin, 6-gingerol, ginsenoside Rb1, atractylode-I) in Lizhong decoction (LZD) extracts were investigated with an in situ single-pass intestinal perfusion model in rats. UPLC-TQ-MS was used to determine the concentration of the five components in the intestinal perfusion. Animal welfare and experimental procedures were in accordance with the regulations of the Animal Ethics Committee of Nanjing University of Chinese Medicine. As evaluation indexes for the intestinal absorption characteristics, the absorption rate constant (Ka) and the apparent permeability coefficient (Peff) of the five main ingredients were analyzed. Results showed that the best absorption sites for glycyrrhizic acid, isoliquiritin and 6-gingerol were the ileum, colon and duodenum, respectively, and the differences between different intestinal segments were statistically significant (P <0.05). There was no notable difference in Ka and Peff between ginsenoside Rb1 and atractylode-I in the different intestinal segments (P > 0.05), suggesting that they were absorbed throughout. The five components were well-absorbed in the whole intestine (Peff > 1.0×10-3 cm·min-1), indicating that LZD is suitable for preparing sustained, controlled release and enteric-coated preparations.

ABSTRACT

Based on the rapid development of COVID-19 epidemic, China has built Huoshenshan Hospital in Wuhan. This paper analyzes the matching degree between drug consumption in Huoshenshan Hospital and medicine prepared by the medical team in the early stage and the use of medicine in the first week after receiving the patients in Huoshenshan Hospital, and compares it with medicine carried by the medical team. With the novel coronavirus infection developing rapidly, the superior group has a tight schedule and a heavy task, and the preparation time before setting out is only 3 days, the matching degree of pharmaceutical supply can reach 60% within a short period of time, which can already meet the general clinical treatment needs. This paper analyzes the situation of preparing medicine in medical teams in the early stage, and provides experience for future medical teams to carry out the law again.

ABSTRACT

OBJECTIVE: To study the improvement effects of Lanqian buccal tablets on scopolamine-induced learning and memory impairment of mice. METHODS: The mice were randomly divided into blank group (normal saline), model group (normal saline), positive group (Donepezil hydrochloride tablets, 1.52 mg/kg) and Lanqian buccal tablets high-dose, medium-dose and low-dose groups (800, 400, 200 mg/kg), with 14 mice in each group. Once a day, 30 days after continuous intragastric administration, except for blank group, other groups were intraperitioneal injected scopolamine hydrobromide 3 mg/kg to induce learning and memory impairment model. After modeling, the learning and memory ability of mice were evaluated with step through test (latency, mistake times of entering darkroom as indexes) and Morris water maze (the time of going up on the platform, the times of crossing the platform, swimming time in target quadrant as indexes). The levels of Ach, ChAT, AchE, SOD and MDA in cerebral tissue of mice were determined. RESULTS: Compared with blank group, latency, the times of crossing the platform, swimming time in target quadrant, the levels of ChAT, Ach and SOD were shortened or decreased significantly in model group (P<0.05 or P<0.01), while mistake times of entering darkroom, the time of going up on the platform, the levels of AchE and MDA were extended or rised significantly (P<0.01). Compared with model group, latency (except for Lanqian buccal tablet low-dose group), the times of crossing the platform, swimming time in target quadrant, the levels of ChAT, Ach and SOD were extended or rised significantly in positive group and Lanqian buccal tablet groups (P<0.05 or P<0.01). The mistake times of entering darkroom (except for Lanqian buccal tablet low-dose group), the time of going up on the platform (except for Lanqian buccal tablet low-dose group) and the levels of AchE and MDA (except for Lanqian buccal tablet medium-dose and low-dose group) were shortened or decreased significantly (P<0.05 or P<0.01). CONCLUSIONS: Lanqian buccal tablet can improve scopolamine-induced learning and memory impairment of mice by increasing the levels of ChAT, Ach and SOD and decreasing the levels of AchE and MDA.

ABSTRACT

Uremic toxins are harmful substances that accumulate in the body when the renal function declines in patients with chronic kidney disease (CKD). It is an important factor contributing to accelerated progression of CKD. There is no effective treatment for reducing uremic toxins. As an extensively used medicine for treatment of CKD in the clinic, Huangkui capsule is effective but the mechanism of its action remains unclear. This study investigated the effect of Huangkui on the accumulation of uremic toxins in CKD rats, with the discussion about its mechanism of action. UPLC-TQ/MS was used to detect the accumulation of uremic toxins in CKD rats after oral gavage with Huangkui. 16S rDNA sequencing technology was used to analyze the gut bacteria composition in rats. HPLC-FLD was used to detect the uremic toxins and their molecular precursors in feces. The effect and mechanism of Huangkui on the uremic toxin precursor in gut bacteria were studied by anaerobic culture system in vitro. All procedures were approved by the Institutional Animal Care and Use Committee of the Nanjing University of Chinese Medicine. The results showed that Huangkui (0.675 g·kg-1) could effectively inhibit the accumulation of uremic toxin indoxyl sulfate (IS) in CKD rats, with IS concentration in rat's plasma, liver and kidney decreased by 49.5%, 68.9% and 40.6%, respectively. Huangkui didn't affect the metabolic pathway of IS in host liver, didn't intervene the process of the IS precursor molecule indole conversion to IS. Instead, Huangkui significantly decreased the indole content in gut, with the indole in CKD rat's feces decreased by 46.4%, suggesting that the gut bacteria may be a target for intervene IS biosynthesis by Huangkui. Huangkui didn't affect the abundance of enterobacteriaceae bacteria (the main gut flora of indole synthesis) in CKD rats, suggesting that Huangkui didn't interfere with indole biosynthesis by directly affecting the abundance of indole synthesis related bacteria. Huangkui at 4 000, 400, 40, and 4 μg·mL-1 showed a dose-dependent inhibition of the indole production by gut bacteria in vitro. The bacteria tryptophan transport concentration decreased from 83.4 μmol·L-1 to 43.6 μmol·L-1 after co-incubated with Huangkui for 12 h, suggesting that Huangkui inhibited indole production of gut bacteria by interfering with tryptophan transportation. These results indicate that gut bacteria may be a potential target for alleviation of uremic toxin accumulation and for delaying CKD progression.

ABSTRACT

Objective To study the cytotoxicity of bi-chimeric antigen receptors modified T lymphocytes (BiCAR-T) on the human acute myeloid leukemia (AML) cell line HL60 in vitro and the anti-tumor effects of BiCAR-T on the NOD SCID mouse model of AML in vivo.Methods The BiCAR-T were prepared and the expression of chimeric antigen receptor (CAR) of prepared BiCAR-T was analyzed by flow cytometry.In vitro study was divided into two groups:the experiment group (BiCAR-T) and the control group (T lymphocyte).The killing rate of BiCAR-T in vitro on HL60 cells was determined by CCK8 assay and the level of interferon-γ (IFN-γ) secreted from BiCAR-T co-culturing with HL60 cells for 48 hours was detected by enzyme linked immunosorbent assay (ELISA) at different effect/target ratios (5∶1,10 ∶ 1,20 ∶ 1).The NOD SCID mice AML model was established by the injection of HL60 cells through tail vein and used to assess the antitumor effects in vivo.The mice were randomly divided into three groups according to the random number table:the blank control group receiving 0.9% NaCl 0.2 ml through tail vein,the model group and the treatment group receiving 1 × 107 HL60 cells in 0.2 ml phosphate buffer saline (PBS).After 20 days,the treatment group was injected with 2 × 107BiCAR-T in 0.2 ml PBS 3 times a week for 2 weeks,while the other two groups received 0.9% NaCl 0.2 ml.The pathological changes in the mice livers and spleens were observed after 2 weeks of treatment.Results The CAR expression rates of BiCAR-T were more than 50.00%.In vitro experiments proved that the killing rates of BiCAR-T in the experimental group and T lymphocytes in the control group on HL60 cells were (25.43 ±1.32)% vs.(16.18 ±0.75)%,(50.33±3.11)% vs.(25.47±1.27)%,and (85.89 ± 3.96) % vs.(49.45 ± 2.77) % at different effect/target ratios (5 ∶ 1,10 ∶ 1,20 ∶ 1).The killing efficiency of BiCAR-T and T lymphocytes on HL60 cells was significantly different (F =404.17,P < 0.001);the killing efficiency of BiCAR-T and T lymphocytes on HL60 cells was significantly different at different effect/ target ratios (F =548.09,P < 0.001);and the killing efficiency on HL60 cells in the experimental group (BiCAR-T) was significantly higher than that in the control group (T lymphocytes) at different effect/target ratios (F =45.36,P < 0.001).The IFN-γlevels secreted from BiCAR-T in the experiment group and T lymphocytes in the control group co-culturing with HL60 ceils after 48 h were (435.65 ± 20.44) pg/ml vs.(356.75 ± 19.87) pg/ml,(1 639.98 ± 95.75) pg/ml vs.(1 109.37 ± 80.98) pg/ml,and (3 467.43 ± 187.54)pg/ml vs.(2 245.52 ± 112.66)pg/ml.The IFN-γlevel in the experiment group (BiCAR-T) and the control group (T lymphocytes) was significantly different (F =156.24,P < 0.001);the IFN-γ level was significantly different at different effect/target ratios (F =857.67,P < 0.001);the IFN-γlevel in the experimental group (BiCAR-T) was significantly higher than that in the control group (T lymphocytes) at different effect/ target ratios of 5 ∶ 1,10 ∶ 1,20 ∶ 1,respectively (F =46.31,P < 0.001).The result of hematoxylineosin staining (HE) staining showed that leukocyte infiltration in the treatment group was significantly decreased compared with the model group.Conclusion The experimental results showed that BiCAR-T is a kind of efficient targeted immunocyte modified by gene engineering,and it can significantly inhibit leukocyte infiltration of AML in vivo and in vitro.

ABSTRACT

The response surface method was used to evaluate the concentration range, property and degree of Scutellaria (HQ) and Coptis (HL) herb pair on improvement of glucose and lipid metabolism in this paper. Type 2 diabetes mellitus (T2DM) was induced in rats by low a dose of streptozotocin (STZ) in combination with high-fat diet feeding. The rats were administered with HQ-HL extract pairs at different ratio (1:0, 3:1, 2:1, 3:2, 1:1, 2:3, 1:2, 1:3, 0:1). To investigate the influence of herb pair extracts on glucose and lipid metabolism of T2DM rats, the multi index synthetic index method was adopted to integrate the indexes, then the response surface method was applied to analyze the integration results. The ratio of HQ and HL from 3:1 to 1:3 showed a synergistic effect (with the value between -0.2 and -0.8). Especially, when the proportion was between 3:2 and 1:3, herb pair extracts possessed stronger synergistic effects (value:-0.8). The high dose of HQ-HL with the ratio of 1:1 exhibited the strongest synergistic effect.

ABSTRACT

Prostate-specific antigen (PSA) is a biomarker for the diagnosis and management of prostate cancer and involved in the development of prostate cancer and/or its progression from the localized to the metastatic stage. This review presents an overview of the roles of PSA in promoting the progression and metastasis of human prostate cancer and its underlying mechanisms, including its serine protease activity, interaction with the cellular membrane receptor, and suppression of specific immune responsiveness, and also points out some of the key problems to be solved.

[Objective]To radiolabel the PSMA aptamer A10-3.2 with 99mTc , and explore its biological characteristics in vivo and in vitro.[Methods]Using Succinimidyl 6-hydrazinonicotinate hydrochloride (SHNH) as the bifunctional chelating agent to label aptamer A10-3.2 with 99mTc, then tested for the stability in vitro, the specific uptake by prostate cancer LNCaP cells (PSMA+) , the characteristics of SPECT/CT imaging and biodistribution in LNCaP tumor-bearing NOD/SCID mice.[Results]The labeling rate and radiochemical purity of the products (99mTc-SHNH-A10-3.2) are(71.31 ± 6.78)% and 97.03%,respectively. 99mTc-SHNH-A10-3.2 had obvious target specificity for PSMA positive prostate cancer LNCaP cells, its uptake rate was significantly higher than PSMA nega?tive PC-3 cells (P<0.01). And in tumor-bearing mice, the tumor has a certain uptake and a high ratio of the tumor tissue to the mus?cle.[Conclusion]This study successfully constructed 99mTc-labeled PSMA-targeted aptamer A10-3.2, which has a good stability and targeting in vivo and in vitro, has a high tumor tissue/muscle ratio in tumor-bearing mice, which show that it may be a potential target?ed molecular imaging agent for prostate cancer.

ABSTRACT

We analyzed the antimicrobial resistance and pulsed field gel electrophoresis (PFGE) patterns of Salmonella strains of infectious diarrhea isolated four hospitals in Shenzhen during 2012-2016.A total of 137 Salmonella strains collected in 4 hospitals in Shenzhen were identified by serotyping,micro-dilution method was used to detect the antimicrobial susceptibility of the isolates.Pulsed field gel-electrophoresis(PFGE) was used for molecular typing and cluster analysis of S.typhimurium and S.enterica strains.Results showed that 137 strains of Salmonella were divided into 30 kinds of serotype.S.typhimurium was the predominant serotype.The drug susceptibility test indicated that the antibiotic resistance of the strains to chloromycetin,ampicillin and amoxicillin were more serious than other drugs,the resistance to penicillins,cephalosporins,aminolycosides and macrolides showed increase trends year by year.Of 49 strains of S.typhimurium were divided 38 molecular patterns by PFGE,with similarity ranged from 65.5% 100%,TY18 was the main PFGE pattern.The 27 strains of S.enterica were divided into 7 molecular patterns by PFGE,with similarity ranged from 61.8%-100%,EN7 was the main PFGE pattern.The status of drug resistance of infectious diarrhea isolates was rather severe,PFGE patterns showed diversity,suggesting the diarrhea cases were distributed sporadically.Partial PFGE patterns and its corresponding antidrug spectrum have certain aggregation relationship.

ABSTRACT

AIM:To clarify whether sulforaphane (SF) has protective effects on retina neuronal cells in dia-betic rats and to identify the related mechanisms involved in this process. METHODS:The diabetic rat model was induced by single intraperitoneal injection of streptozotocin (STZ). The protective effects of SF were evaluated by measuring the generation of reactive oxygen species(ROS),detecting apoptosis of retina neuronal cells with TUNEL staining and counting the survival retinal ganglion cells (RGCs). The nuclear translocation of nuclear factor E2-related factor 2 (Nrf2) and the protein expression of heme oxygenase-1 (HO-1) were examined by immunofluorescence analysis and Western blot. RE-SULTS:SF treatment significantly attenuated ROS generation, decreased the apoptosis of retina neuronal cells and in-creased the numbers of survival RGCs in the diabetic rats. Meanwhile,SF significantly increased the nuclear accumulation of Nrf2 and the protein level of HO-1 in the retinas of diabetic rats. However,HO-1 inhibitor,protoporphyrin Ⅸ zinc(Ⅱ) diminished the inhibitory effects of SF on RGCs apoptosis. CONCLUSION:SF partially exerts the beneficial neuroprotec-tive effects via the activation of the Nrf2/HO-1 antioxidant pathway,therefore alleviating retinal oxidative stress and decrea-sing the apoptosis of retina neuronal cells.

ABSTRACT

Objective To investigate the long-term efficacy of calcium phosphate cement combined with bFGF in repairing pulp chamber perforation and to analyze the correlation between the diameter of the perforation and the curative effect.Methods 75 patients with pulp chamber perforation (82 teeth) were enrolled and divided into the observation group and the control group according to the repair material and method.A series of subgroups were also modeled according to the diameter of the perforation,which include the control group A (≤ 1.5 mm),control group B (1.6~3 mm) and control group C (>3 mm),as well as the observation group A (≤ 1.5 mm),observation group B (1.6~3 rmm) and observation group C (>3 mm).The observation group was treated with calcium phosphate cement combined with bFGF,and the control group was treated with calcium phosphate cement alone.Results The total effective rate of the observation group was 97.8%,which was significant higher than 80.6% in the control group (P<0.05).The cure rate of the observation group A was 100%,which was significant higher than 73.3% in the observation group B and 41.7% in the observation group C (all P<0.05).The total effective rates of the observation group A and B were significantly higher than 91.7% in the observation group C (all P<0.05).The cure rate of the control group A was 92.9%,which was significant higher than 60.0% in the control group B and 25.0% in the control group C (all P<0.05).The total effective rates of the control group A (100%) and B (90.0%) were significantly higher than 91.7% in the control group C (all P<0.05).Conclusions Calcium phosphate cement combined with bFGF in repairing the pulp chamber perforation was significantly better than calcium phosphate cement alone.The cure rate of perforation repairing is closely related to the perforation size.The perforation with small diameter may achieve a better repairing effect.

ABSTRACT

The roots of Paeoniae lactiflora Pall. are widely consumed as crude drugs in Asian countries due to their remarkable beneficial health effects. The present research was undertaken to illuminate the dynamic changes in metabolites and enzymes and facilitate selection of the harvesting time when the herb can provide optimum health benefits. P. lactiflora roots were analyzed at 12 stages of growth for monoterpenoid glycosides, phenols, nucleosides, nucleobases, amino acids, and polysaccharides by high-performance liquid chromatography with photodiode array detector, ultra-high pressure liquid chromatography coupled with tandem mass spectrometry, and UV spectrophotometry. The enzyme activities of plant β-glucosidases and esterases were determined by UV methods. The total content of monoterpenoid glycosides and phenols peaked in December. For nucleosides and nucleobases, the highest content appeared in April. The maximum phasic accumulation of the total amino acids took place in March, and the content of total polysaccharides reached a peak value in September. December, April, and March were selected as the appropriate harvesting times for producing natural medicinal or health food products. Plant β-glucosidases and esterases showed the highest activity in December and May, respectively. When the activity of β-glucosidase increased, esterase activity decreased, while the contents of oxypaeoniflora and paeoniflorin increased. When esterase activity increased, the contents of benzoylpaeoniflorin, paeoniflorin, and gallic acid decreased. In conclusion, the results from the present study would be useful in determination of the suitable time for harvesting P. lactiflora roots for medicinal purposes.

BACKGROUND:Parathyroid hormone can activate a series of physiological and biochemical reactions through specific receptors on the surface of target organs,to adjust the dynamic calcium balance.OBJECTIVE:To explore the mechanisms whereby parathyroid hormone maintains in vitro proliferation of bone marrow mesenchymal stem cells (BMSCs),by observing the effects of parathyroid hormone on calcium ions in BMSCs.METHODS:BMSCs at passage 4 were randomly divided into two groups.Experimental group was treated with parathyroid hormone,and control group was treated with routine culture medium.During 10 days of culture,MTT assay was used to detect BMSCs proliferation.At 7 and 14 days after treatment,laser scanning confocal microscopy was used to detect fluorescence intensity of calcium ions in BMSCs in the two groups.RESULTS AND CONCLUSION:(1) At 1 and 2 days after culture,the cell viability in the two groups was sluggish.At 3 days after culture,the cells turned into the rapid growth.At 7 and 8 days after culture,the cell growth turned into the plateau.At 10 days after culture,the cell proliferation ability had a weakening trend,which was lower in the control group than in the experimental group (P < 0.05).(2) At 7 days after culture,the fluorescence intensity of intracellular calcium ions was strong.As the induction time extended,the fluorescence intensity of intracellular calcium ions was declined at 14 days after-culture.At 7 and 14 days after culture,the fluorescence intensity of intracellular calcium ions in the experimental group was higher than that in the control group (P < 0.001).There was no difference in the fluorescence intensity of intracellular calcium ions between experimental group at 14 days and control group at 7 days.To conclude,parathyroid hormone could increase the fluorescence intensity of calcium ions in BMSCs,and function to maintain the BMSCs proliferation.

Objective To observe the effect of bandage contact lens on cornea epithelial healing after Epi-LASIK,and to evaluate the efficacy and safety.Methods Prospective case-control study.The 92 cases(181 eyes) performing Epi-LASIK were devided into observation group and control group.The 46 cases(91 eyes) of the control group were covered with routine nursing and the other 46 cases (90 eyes) of the observation group were served with bandage contact lens after Epi-LASIK.The symptoms and signs,time of corneal epithelial recovery,visual acuity and pain of postoperation of the two groups were examined within one week.Results After Epi-LASIK treatment,there was no significant difference in visual acuity between the two groups(P > 0.05).At the 1 st,3rd,and 7th day after Epi-LASIK treatment,the ache grading results of the observation group were (2.53 ± 0.62),(1.39 ± 0.71),(0.07-± 0.25);and the ache grading results of control group were (3.22 ± 0.79),(1.74 ± 0.49),(0.26 ± 0.49).Pain scores of observation group were significantly lighter than that of the control group (P < 0.05).After Epi-LASIK treatment,the average time of corneal wound healing in the observation group and control group were (2.80 ±0.86) days and (4.07 ±0.68) days respectively.The average time of postoperative corneal wound healing in the observation group was shorter than control group(P < 0.05).Conclusion The application of bandage contact lens on cornea epithelial healing after Epi-LASIK treatment can reduce the pain of patients and promote the recovery of cornea in myopia patients.

ABSTRACT

<p><b>OBJECTIVE</b>To examine the incidence of congenital heart disease (CHD) in children aged 0-3 years in the rural areas of Chongqing, and to determine the suitable "screening-diagnosis-follow-up" system and screening indicators for CHD in these areas.</p><p><b>METHODS</b>Children aged 0-3 years from rural areas of the Fuling Disctrict of Chongqing were selected by cluster sampling. Using the "screening-diagnosis-evaluation system" employed at the levels of village/town, district/county, and province/city, the children were screened for seven indicators, i.e., family history of CHD, dyspnea, cyanosis, unique facial features, other congenital malformations, heart murmurs, and blood oxygen saturation (SpO<95%). Children who were positive for one or more indicators accepted echocardiography (ECG) for the diagnosis of CHD. CHD patients were evaluated for disease progression, given guided treatments, and followed-up by pediatric cardiologists.</p><p><b>RESULTS</b>Screening was performed for 10 005 out of the 10 281 children enrolled in the study (97.32% response rate). Among the 175 children who were positive for the indicators, 166 underwent ECG and 60 (0.6‰) were diagnosed with CHD, including 46 cases of simple CHD (76.65%), 11 cases of combined CHD (18.33%), and 3 cases of complex CHD (5.00%). Of the 7 screening indicators, heart murmur had the largest area under the ROC curve for the diagnosis of CHD. In addition, a combination of screening indicators (heart murmur, unique facial features, and other congenital malformations) was most effective for screening out CHD. The CHD patients were given surgical or intervention treatments, and followed up for 6 to 18 months. Ten patients improved without treatment, 13 patients received interventional or surgical treatment, 1 patient died of non-cardiac reasons. The remaining 36 patients were subjected to further follow-up.</p><p><b>CONCLUSIONS</b>Heart murmur alone and in combination with unique facial features and other congenital malformations are valuable tools for CHD screening in children aged 0-3 years. The "village/town-district/county-province/city" screening-diagnosis-evaluation systems are useful for the early detection, diagnosis, and treatment of CHD in infants and young children from the rural areas of Chongqing.</p>

ABSTRACT

Objective  To investigate the optimized range of body mass index (BMI) selection for patients undergoing 70 kV high-pitch dual-source coronary CT angiography (CCTA) on the third-generation dual-source CT (DSCT). Methods  Patients undergoing prospective high-pitch ultra-low contrast media (CM) CCTA on the third-generation DSCT using the automatic tube voltage selection at 70 kV were included and divided into three groups:group A,with BMI≤24 kg/m;group B,with 24 kg/m<BMI<28 kg/m;and group C,with BMI≥28 kg/m. The baseline data,image quality results,and effective dose (ED) were evaluated and compared. Results  No significant difference was found in age (H=2.36,P=0.78),heart rate (H=3.51,P=0.80),and ED(H=1.77,P=0.12) among three groups. The noise of the aortic root in group A was (21.66±4.00)HU,which was significantly lower than that in group C [(23.61±4.20)HU](F=2.05,P=0.03). The signal to noise ratio of aortic root in group A (18.61±5.28) was significantly better than that in group C (14.75±2.58) (F=5.31,P=0.02). No other significant difference in quantitative image quality were found among the three groups (all P >0.05). Subjective image quality in group A(Z=2.91,P=0.004) and B(Z=2.27,P=0.021) were both significantly better than that in group C. Conclusion  The ultra-low tube voltage (70 kV) combined with ultra-low CM CCTA protocol on third-generation high-pitch DSCT may be better for patients with BMI<28 kg/m2 than those with BMI ≥28 kg/min China.

Salviae Miltiorrhizae Radix et Rhizoma residues were pre-treated with acid and alkali, degraded by using cellulose, and the effects of different processing methods on the extraction rate of tanshinones were compared to provide scientific basis for development and utilization of tanshinones from Salviae Miltiorrhizae Radix et Rhizoma residues. The results showed that in the Salviae Miltiorrhizae Radix et Rhizoma residues without pre-treatment, enzymatic hydrolysis time of 4.5 d could make most of the cellulose degraded when the concentration of substrate enzyme concentration was 6 U•mL-1, and the highest glucose concentration was 59.74 mg•g⁻¹. It was found that the best effect was achieved after alkali pre-treatment-cellulose C degradation among the different pre-treatment methods, and the glucose content reached 119.50 mg•g⁻¹, followed by the same concentration of acid pre-treatment-cellulose C degradation. The extraction amount of tanshinone ⅡA was increased by 82.54% after enzyme degradation, with a mass fraction of 2.451 mg•g⁻¹; extraction amount of tanshinone I was increased by 81.82% after enzyme degradation, with a mass fraction of 2.373 mg•g⁻¹; extraction amount of cryptotanshinone was increased by 64.4% after enzyme degradation, with a mass fraction of 1.080 mg•g⁻¹; extraction amount of dihydrotanshinone I was increased by 61.3% after enzyme degradation, with a mass fraction of 0.601 2 mg•g⁻¹. Acid and alkali pre-treatment combined with cellulose degradation could effectively improve the extraction rate of tanshinones from Salviae Miltiorrhizae Radix et Rhizoma residues. This method is operable and practical, and it is beneficial for improving the utilization efficiency of tanshinones (resource based chemicals) from Salviae Miltiorrhizae Radix et Rhizoma residues.
